Dont panic and take plenty of test drives. I test drove all the A6 versions before choosing the 550. No contest. That said, the S6 is now out and reportedly its a real performer and only a few K more than the BMW. As for the Merc, I didn't test any as I just don't like em. Styling makes me sleepy. I think Edmunds tested the A6, E550 and 550, and the 550 easily bested them.

My Ride: 2005 545i with everything
The current E-class is dated and due for replacement in the next 18 months. Buy one now and it will be yesterday's news with half of your lease still to go. It's also considered Mercedes least desired recent model. It just looks really plain IMO and I am a fan of previous E-class V8 models.
The audi A6 is a great car but nowhere near the 550i in terms of performance. Now the S6 is a wickedly fast and awesome car but extremely expensive, as in range of the M5 or higher with a detuned Lambo V10.
Now if I ever saw snow the Audi would be more appealing to me.

I never even considered anything else. This car on order will be my 10th BMW. As long as I can afford it, I'll always have a couple of them. (Ofcourse, if I have one, Mrs. poleposition has to have one as well) There are definitely other nice cars out there but none with the same personality or presence of a BMW. They fit our (my family) personalities and lifestyles to a tee. This wont help you make your decision but I think it's the kind of decision you have to make on your own.

I had the same decision to make...stepping out of an E60. From a design standpoint, if you go Audi, the A5 I think, is a fresh, modern design - though two-door. (Interior finishes are also superior to the "feel" of Merc or BMW.) Also, the A6 design is nearing the end of its cycle however, the '08 S4 is quite a bit larger than the previous 4, and also pretty fresh-looking. Same with the Merc. The E-Class is getting long-in-the-tooth, though the AMG is pretty powerful! The C class (Sport seden), has a fresher look and I think better designed than the S-Class. I considered getting an AMG, a 650 convertible, or a 550 to replace my 545, but decided to go with the 335 convertible because the design is fairly new, twin-turbo, and cool, hardtop, convertible action! I guess it really boils down to your car priorities: size/# of doors, speed, power, design, technology, handling, brand...whatever floats your boat! Good luck with your decision!

I've never owned an A6 but I did test drive one. The seats are better than the 5 series standard / sport ones but the suspension is dreadful. It feels a cheap ride.
I have owned an E Class. The ride quality is superb and, IMO it looks and feels a more upmarket car than the 5 series. However, I had real problems with the drivers seating position (leg ache) and had numerous electrical glitches (it was a brand new car as well). I got rid of it after 4 months.
Personally, I think that the 5 Series is best car, as long as you go for comfort seats, standard suspension and resist the temptation to upsize the alloys.
