Key fob comfort access question?

Hi guys and gals, newbie on this site! 07 525i sports pkg.
Everything was working locks, unlock, trunk , etc. A while back I disconnect the battery so not sure if that had anything to do with it, but now I have to put the key in the column slot to start it, the doors still unlock remotely and trunk, but cannot lock at all, not even the finger on the keyhole trick that used to work. I changed the fob battery and tried some of the programming steps I’ve seen. I’m still thinking it’s a programming issue.
Also the key in the fob, does it actually turn the cylinder? Mine does not and not even sure if it’s supposed to, the key inserts fine, but nothing actually turn to the left or right on the door or trunk.
thanks in advance if I can just get it to lock I’d be happy with that. I’m thinking I’m not performing the right programming method.
