I think all the usual suspects have been listed. In my mind however, there is one "competitor" that hasn't been mentioned... the 3er!
From personal experience, and from what I read on forums, a number of people ultimately choose between a 3er or 5er, depending on needs and type of drive they are after. (No-one said it couldn't be another BMW!)

Agreed, its like comparing a Honda civic to an Accord. E class without a doubt is the closest competitior. E320 is roughly the same in specs with the 6 cylinder E60 and the 545/550 with the E500.

I think the E Class is no longer a competitor and in fact its now the CLS. Before I thought the previous W210 MB was a solid competitor but that was only because it was going against e39. The rest of the "competiton" just tries to come close to the E60 in terms of technology and styling. The only thing I can think of that competes is the CLS. New E Class feels and looks tiny.
+1 The CLS is MB's attempt to take the style and refinement of the E-class and mate it with more taut and punchy delivery. That makes it, to my mind, the closest competitor to the e60.

To me the biggest competitors are:
Mercedes-Benz E-class sedan
Lexus GS
Infiniti G35 sedan
You might think I am crazy to even put G35 sedan there instead of M35/M45, but I really think G is closer to the 5 than M is. I only want to put RWD sedans there, so Acura RL and Audi A6 would be eliminated by me immediately.

You definitely have to include the A6 as a competitor. But only in Quattro, a front wheel drive Audi is worthless.
