Is it possible to cancel guidance without turing the car off once you have started guidance? Thanks in advance.

Yes you can. Just click on the address that you want to go earlier and it will cancel it out. Sorry it's kinda hard for me to explain it in words.
Highlight the address that is currently in use, click on the iDrive button, and scroll to "delete it". Or you can just hold down the nav voice button on the steering wheel (unless you've programmed it for something else) and the voice will go away. I do the latter when I don't want to delete something I may have in my address book but want to "cancel" the nav.

To cancel guidance to a destination (without simply muting it or deleting the destination completely) you click on the destination and uncheck the box that says something like "start guidance." (If you just mute the voice, the HUD will still display arrows, etc.)
It's that simple...
It's that simple...
Here's what I do:
1) Issue voice command "Last Destinations"
2) Turn iDrive knob one 'click' to the right. (This will put it on the current destination.)
3) Push down on the iDrive controller. This selects the current destination.
4) Push down on the iDrive button. Guidance is cancelled. What this does is select the 'start guidance' item, which is in red, if it's the current destination.
No looking at iDrive is necessary; it's very easy.

My Ride: 2006 550i Sport Package, Cold Weather Package, Premium Sound Package, Comfort Access, Xenon Headlights, Adaptive Headlight Control, Navigation System, Steptronic Transmission. Silver Gray Metallic with Gray Leather. Added Red rear reflectors. Swapped stock Style 124 wheels and Bridgestone run-flat tires for BMW Style 125 5-Double Spoke wheels, 8" x 19" front, 9.5" x 19" rear with Michelin Pilot Sport PS2 tires, 245/40YR-19 front and 275/30YR-19 rear.
There is an Icon to the left of the main map. Push iDrive knob to left once, scroll to Icon that shows arrow pointing at a dot and press iDrive knob down. Destination is cancelled.
