No, the sprintbooster is available for all transmissions. For the step and SMG, you need to order the auto version and for the 6speed, there is a manual version available. If you use the wrong one or the pins are a little bent, then you will get a service engine soon or something light come up on the dash. if you are the fence of purchasing one, put it on and drive around and then take it off the car and you will notice a night and day difference. I was going to sell mine but now I can't live without it.
Interesting, you are correct on that. I was told that the SB for manuals are way too sensitive so people recommend only using the SB on Autos/Steps/SMGs. I don't see why a manual would need one......... But there is a significant difference in the increase of throttle response and shifting response with the SB installed.

The short answer is it makes the throttle response much faster. So far, my warranty is still alive and well (and they even kept the car for 3 days - sprint booster and all).
