Is the ventilation very subtle (what I feel in my car even at maximum level 3) or the air flow should be really noticeble?
Probably I have to check mine at the garage?

Its quite subtle, but very effective.
When the cooling is on max, you should hear the seat fans working - you have to have the engine running...if no fan noise then get the dealer to check it.

You should hear the fan on the highest level but the movement of air is very subtle. You should really notice the difference on the hot humid days after being in the car for a few minutes

Question for your seat ventilations... does it blow air thoughout the chair? Like including the back of the buns, lower back and upper back? or is it just like the front of your thighs?

The ventilation is through the seat base and through the seat back.
Its an option on the comfort seats

How does Lexus's seat ventillation differ?
Btw you actually don't need to have Comfort seats any more, this changed for '06 production, you still nead Nasca leather & the new required option of Premium package.
Peace
